Assessment of the dynamics of some cardiovascular and respiratory systems indicators and lung histopathology after administration of baclofen and its combination with ethanol
Backgraund: Baclofen is a selective GABAB agonist, which has a pronounced psychotropic effect and is often the subject to abuse. The effect of baclofen on the cardiovascular and respiratory systems has not been studied. Aims: The aim of the study was to assess the dynamics of some functional parameters of the cardiovascular and respiratory systems and lung histopathology in baclofen and baclofen-ethanol poisoning. Materials and methods: Balofen and its combination with ethanol were administered to rats under general anesthesia (chloral hydrate). The controls (n=5) were not treated to baclofen or ethanol. Group 1, 3, 5 animals were treated to baclofen (85 mg/kg), physiological parameters were recorded in 3, 4.5 and 24 hours. Groups 2, 4, 6 animals were treated to baclofen (85 mg/kg) and 40% (vol.) ethanol (7 ml/kg). The saturation, RBCs number (per liter), hemoglobin (g/l) were measured. Then, the animals were euthanized by the displacement of the cervical vertebrae biological material was taken for following studies. We performed morphometric analysis using the grid of G.G. Avtandilov. Results: As a result of baclofen and baclofen-ethanol administration, the saturation decreased. RBC number (per liter) and hemoglobin level (g/l) remained unchanged. 3 h. after baclofen-ethanol administration, an increase in the share of the vessels was noted. 4,5 hours after baclofen- ethanol administration, an increase in the share of WBCs, the ratio of the share of WBCs to the area of the interalveolar septa (IAS), the share of edema was noted. 24 h. after baclofen and baclofen-ethanol administration, the share of edema, WBCs, the ratio of the share of WBCs to the area of IAS, increased. 4,5 hours after baclofen-ethanol administration, the share of WBCs increased. Conclusions: The data obtained as a result of our research with the results of a chemical study, allows us to establish the fact of baclofen poisoning, the time elapsed since the poisoning and the conditions of the drug action. Understanding the processes occurring in the lungs after administration of baclofen and its combination with ethanol will help to provide more effective medical care to this category of patients.
