Features of restructuring chromatin and changing the level of relative expression of genes IL8, IL-1β AND TNFα of neutrophilic granulocytes influenced by glucose-aminyl- muramyl-dipeptide and interferon-γ in patients with chronic sinusitis in vitro

The simultaneous study of induced relative (induced/spontaneous) restructuring of chromatin (RC) and the relative expression of genes IL8, IL-1β and TNFα of peripheral blood neutrophil granulocytes (NG) in healthy persons and in patients suffering chronic sinusitis with frequent exacerbations at acute stage (clinical model of immunodeficiency) was carried out in vitro. To evaluate a level of spontaneous and induced RC the author's method was applied using at final stage a polarization microscope with crossed analyzer and polarizer, and to detect IL8, IL-1β and TNFα genes relative expression a polymerase chain reaction was used. It was shown that in healthy persons the levels of relative (induced/spontaneous) RC were higher than in ill ones; and the induction with glucose-aminyl-muramyl-dipeptide (GMDP) as well as interferon-γ (IFNγ) caused a significant increase of relative NG RC, followed by IL8, IL-1β and TNFα genes expression in comparison to non-induced control. A disturbance in relative NG RC levels was shown in the patients with chronic sinusitis at acute stage and it was tightly associated with the decrease in IL8, IL-1β and especially TNFα genes relative expression. Presumably the study of relative (induce/spontaneous) NG RC may be used at screening examinations as an integral method which will allow to suspect the disturbances in NG genes expression in patients suffering immunodeficiency with infectious syndrome, stipulated by NG functioning defects.
