Phenotypical Transformations of IFN alpha/beta R1(+)IFN gamma R(+)TLR4(+) Subpopulation of Neutrophilic Granulocytes in Chronic Herpes-Viral Infections and their Experimental Modulation by Recombinant IFN alpha 2b

The experimental model of the in vitro transformed phenotype of neutrophilic granulocytes (NGs) equipped with receptors IFN alpha beta R1, IFN gamma R, TLR4 of patients with a variety of chronic herpes viral infections (ChHVI), and capability to modulate negative altered phenotype of IFN alpha beta R1(+)IFN gamma R(+)TLR4(+) NGs subpopulation of the peripheral blood of these patients under the influence of recombinant IFN alpha 2b in the system in vitro. The features of the transformed phenotype of NGs were described. The influence of recombinant IFN alpha 2b on untransformed and transformed phenotype of NG was investigated in vitro. It was shown that in group's patients with ChHVI, the percent of NGs, expressing IFN alpha beta R1 and IFN gamma R increased compared with the control group of conditionally healthy individuals. In this, the number of TLR4+ NGs of these patients was divided into two groups with high reliability of differences between the groups and differed from the control group. In accordance with this, in one group the number of NGs expressing TLR4 receptors increased in comparison with the control group of conditionally healthy individuals, and in the other group - decreased. In group's patients with ChHVI, under the influence of recombinant IFN alpha 2b the number of NGs, expressing on the surface membrane all receptors IFN alpha beta R1, IFN gamma R and TLR4, was decreased. In addition, IFN alpha 2b had greatly increased the density of IFN alpha beta R1(+) NGs, but moderately reduced the density' expression of IFN gamma R+ NGs of group's patients with ChHVI.
